Morfogénesis in vitro de leguminosas forestales nativas de la República Argentina

Resumen

El objetivo de este trabajo fue inducir la morfogénesis in vitro en cotiledones inmaduros de leguminosas forestales nativas de la República Argentina: Erythrina crista-galli L. (seibo), Acacia caven (Mol.) Mol. (espinillo), Parkinsonia aculeata L. (cina-cina), Gleditsia amorphoides (Griseb.) Taub. (espina de corona) y Sesbania punicea (Cav.) Benth. (acacia mansa). Las legumbres verdes fueron recolectadas de árboles que crecen naturalmente en los alrededores de la ciudad de La Plata (34º 55´ S; 57º 57’ W; 15 m snm), Provincia de Buenos Aires. Los frutos fueron desinfectados en superficie, se extrajeron las semillas inmaduras y se separaron los cotiledones del eje embrionario. Los cotiledones fueron cultivados en el medio basal Murashige-Skoog (MS), con sus macro y micronutrientes a la mitad de su concentración, suplementado con 3% de sacarosa, 0,65% de agar, 0,165 g/l de NO3NH4, 1 ppm de 2,4-D (ácido 2,4- diclorofenoxi acético) y 0,1 ppm de Kin (cinetina). Los cultivos fueron incubados a 25 +/- 2 ºC y 16 horas de luz con una irradiancia de 60 mmol m-2 s-1. Se obtuvieron embriones somáticos en Acacia caven, regeneración de brotes y raíces adventicias en Parkinsonia aculeata, callos y raíces en Gleditsia amorphoides y callos en Erythrina crista-galli y Sesbania punicea. Los resultados sugieren que algunas leguminosas nativas de la República Argentina pueden ser propagadas mediante técnicas de cultivo de tejidos.
